In-depth explanation of DNA Replication

DNA replication copies the double helix so that each daughter molecule contains one original strand and one newly synthesised strand, a mechanism known as semi-conservative replication. Helicase unwinds the double helix at the replication fork, and single-strand binding proteins keep the strands apart. Primase synthesises short RNA primers, and DNA polymerase adds nucleotides only in the 5-prime to 3-prime direction. The leading strand is made continuously, while the lagging strand is made as Okazaki fragments that ligase later joins together. Proofreading by DNA polymerase brings the overall error rate to roughly one mistake in a billion nucleotides.

Going deeper: DNA Replication

Key ideas: Semiconservative: each daughter has one old and one new strand. Helicase unwinds, single-strand binding proteins stabilize, primase lays an RNA primer, DNA polymerase III extends 5 to 3 only, giving a continuous leading strand and discontinuous Okazaki fragments on the lagging strand, joined by ligase after polymerase I replaces primers. Error rate is about 1 in 10 to the 9 after proofreading.

Common mistakes and connections: Students think polymerase builds in both directions or that replication starts anywhere randomly; it begins at origins. Another error is mixing up leading and lagging strands. Links to PCR, telomeres, mutation and cell cycle.

Visualization of DNA Replication

After two generations in light nitrogen, half of the DNA is hybrid and half is light, the pattern that proved replication is semi-conservative. The interactive lesson shows this as a live animation or chart.

Worked example and use case

The Meselson and Stahl experiment with nitrogen isotopes proved that replication is semi-conservative.
