Heterocyclic Compounds

Rings containing atoms other than carbon, such as pyridine, furan and pyrrole.
Heterocyclic compounds are cyclic molecules in which one or more ring atoms are elements other than carbon, most commonly nitrogen, oxygen or sulfur. Many heterocycles are aromatic, such as pyridine, pyrrole, furan, thiophene and imidazole, and their heteroatoms affect basicity, reactivity and hydrogen bonding. Heterocycles are extremely common in nature and medicine, appearing in nucleic acid bases, vitamins, alkaloids and most modern drugs. Their chemistry is a central part of pharmaceutical design.
Key ideas: Rings containing at least one non-carbon atom (N, O, S). Five-membered aromatic: pyrrole, furan, thiophene (heteroatom lone pair is part of the 6 pi electrons, so electrophilic substitution occurs at C2). Six-membered: pyridine, pyrimidine. Reactivity toward electrophiles: pyrrole > furan > thiophene > benzene > pyridine. Appear in nucleic acid bases, vitamins, and many drugs.
Common mistakes and connections: Students claim pyrrole is basic like pyridine; its lone pair is used in aromaticity, so it is not. Another error is writing the wrong site of substitution, C3 rather than C2. Links to aromaticity, pyridine, alkaloids and pharmaceuticals.
Heteroaromatic rings are aromatic but less stabilized than benzene; the larger the lone-pair donor strain, the lower the resonance energy. The interactive lesson shows this as a live animation or chart.
The purine and pyrimidine bases in DNA are nitrogen-containing heterocycles.
